Scientists use light to 'tag and track' genetic processes

In a new study, UT Dallas researchers outline how they used fluorescent molecules to "tag" DNA and monitor a process called DNA looping, a natural biological mechanism involved in rearranging genetic material in some types of cells.

The UT Dallas "tag and track" method not only sheds light on how DNA loops form, but also might be adapted to screen drugs for effectiveness against certain viruses that shuffle genetic material, such as HIV.

Until now, scientists primarily had "snapshots" of the initial and final stages of DNA loop formation, with only limited information about what happens during the intermediate steps, said Dr. Stephen Levene, professor of bioengineering, molecular and cell biology, and phyiscs at UT Dallas. He is senior author of the study, published online and in an upcoming issue of the journal Nucleic Acids Research.

"Scientists have known for more than 30 years that DNA looping is an important part of molecular biology and gene regulation, but until our work, there have been few serious attempts to understand the basic biophysics of the process," Levene said.

DNA looping is a mechanism common in many instances of natural gene-splicing. Proteins within cells or proteins made by invading viruses latch onto specific docking points on a DNA molecule. They bring those points together to form a loop, and then snip out the genetic material between the points while reconnecting the now-loose ends.

DNA loop formation is especially important in organisms whose genetic material is circular, including some bacteria and viruses. Human DNA is linear, but the possibility that DNA looping takes place in human cells is an ongoing area of investigation, Levene said.

Levene and UT Dallas doctoral student Massa Shoura, the lead author of the paper, used a protein called Cre in their experiments. Cre is made by a virus that infects bacteria and is so good at forming DNA loops and excising genetic material that scientists routinely use it to delete genes from laboratory animals, which are then used to study the role of genes in human disease.

Levene and Shoura engineered isolated segments of DNA to contain Cre's docking points. They also inserted into those points a molecule that fluoresces when exposed to certain wavelengths of light. By monitoring the changes in fluorescence, the researchers could watch the steps of the loop formation.

The information the researchers have gleaned is not only useful for understanding basic biology and genetics, but also might lead to more efficient methods for screening potential new drugs for anti-HIV activity.

Once inside a host cell, HIV produces an enzyme similar to Cre, called an integrase. As its name suggests, the integrase slices into the host's DNA and inserts HIV's genetic material.

"Our fluorescent-tag technique could be used in the lab to more closely examine how HIV inserts itself into the host's genome," Shoura said. "By labeling and monitoring the process, we also could test drugs designed to interfere with the integrase."

"We estimate that using fluorescence-based methods such as this for drug screening could be as much as 10,000 times more efficient than methods that are currently used," Levene said.

Other UT Dallas researchers from the Department of Molecular and Cell Biology who participated in the study were senior scientist Dr. Alexandre Vetcher; doctoral students Stefan Giovan, Farah Bardai and Anusha Bharadwaj; and former undergraduate student Matthew Kesinger. The National Institutes of Health and the National Science Foundation funded the research.

Wall Street dips after six-day rally; Japan data weighs

NEW YORK (Reuters) - Stocks fell on Monday after six days of gains for the S&P 500 and as Japanese data showed their economy grew much less than expected in the second quarter, a reminder of the headwinds faced by the global economy.

The benchmark index had risen 3 percent over the prior six sessions, its longest rally since December 2010, but gains have slowed, with the index hovering at highs not seen since May. The S&P 500 was still up 1.6 percent for the month.

"Coming off the heels of a light week on the economic front, the markets remain in what some have called a state of 'melting up.' Whether the market's strength is artificially induced or not, the greater risk for traders at the moment is in missing out on the upside," said Randy Frederick, Managing Director of Active Trading & Derivatives at Charles Schwab.

With "the European Central Bank, the U.S. Federal Reserve and the People's Bank of China all standing in the wings ready to take action to support their respective economies, the downside risks in the market have been greatly reduced at the moment," he added.

The Dow Jones industrial average was down 65.34 points, or 0.49 percent, at 13,142.61. The Standard & Poor's 500 Index was down 5.24 points, or 0.37 percent, at 1,400.63. The Nasdaq Composite Index was down 6.70 points, or 0.22 percent, at 3,014.16.

Japan's gross domestic product expanded just 0.3 percent in the April-June period, half the pace expected, raising doubts about the strength of the recovery and highlighting the impact of Europe's debt crisis on worldwide demand.

But the CBOE VIX Volatility index , which is seen as a proxy for investors' fears, fell 3.5 percent to 14.22. The decline was unusual as the index typically moves inversely to the S&P 500, suggesting investors were not overly concerned about the market's outlook.

Late on Friday, the head of the San Francisco Federal Reserve said the Fed should launch a fresh round of bond buying to lower the U.S. unemployment rate more quickly, fueling speculation that the central bank could soon unveil a new round of quantitative easing.

Much of the money entering the U.S. equity market since the rally started in June has gone into defensive sectors. But investors are eyeing early signs that more aggressive areas of the market -- such as small-caps and cyclical sectors -- are starting to do better, a key factor if the rally is to continue.

"Those are the sectors that will need to lead to continue the rally, whether they can do it on their own remains to be seen. I'm not sure we will get out of the summer without a pullback," said Janna Sampson, co-chief investment officer at OakBrook Investments LLC in Lisle, Illinois.

Sampson said she was still cautious over the potential for developments in Europe's debt crisis to blindside the market, adding she would be closely watching data releases to see if improvement in the U.S. labor market would continue.

"It is encouraging that offensive areas of the market are joining the rally," wrote Ari Wald, a technical analyst at Brown Brothers Harriman in New York. "In particular, the Russell 2000 , the S&P 500 High-Beta Index , and the iShares MSCI EAFE Index , broke above key resistance last week and completed near-term bottoming formations."

"This 'risk-on' breakout should provide a tailwind for the market and assist the S&P 500 in its attempt to move above its 1,420 April cycle high," he said. "A failure to confirm new cycle highs in the coming weeks to months would create worrisome multimonth divergences."

Tesoro Corp moved on Monday to create the biggest U.S. refining empire in the Pacific Basin with a $2.5 billion deal to buy BP's Carson plant in the isolated California market. Shares in Tesoro rose 8.8 percent to $38.64 while U.S.-listed shares of BP fell 0.5 percent to $42.17.

According to Thomson Reuters data through Monday, of the 454 companies in the S&P 500 that have reported second-quarter earnings to date 68 percent have reported earnings above analyst expectations, matching the beat rate for the last four quarters.

Fatigued Wall Street ends flat on low volume

NEW YORK (Reuters) - Stocks ended little changed on Tuesday in what investors described as a fatigued market after the S&P 500 rose in seven of the past eight sessions.

Retailers were a notable standout after Home Depot raised its profit forecast due to improvement in the housing market and U.S. retail sales data that was stronger than expected, though earlier figures were revised lower. The S&P Retail Index rose 0.9 percent, giving an early boost to the market.

But with the S&P 500 up 12 percent so far this year, gains came off by the afternoon. In the last five sessions, the S&P has not moved more than 0.22 percent in either direction and volume has declined in what is normally a seasonally slow period.

"For the past week or two, the market rally has been largely driven by the absence of sellers and complacency that things will be OK until the end of the month, at least," said James Dailey, portfolio manager at TEAM Asset Strategy fund in Harrisburg, Pennsylvania.

The S&P 500 is within a stone's throw of new four-year highs, leaving investors looking for new catalysts to move the market higher. Just 5.16 billion shares changed hands on the three major U.S. exchanges, a light day of trading.

Home Depot Inc , the world's largest home improvement chain, gained 3.6 percent to $54.71 and gave the Dow the greatest boost after the company reported a higher-than-expected quarterly profit and kept a tight lid on costs to offset weakness in sales.

Home Depot is seeing improvements in California and Florida, states that were hit hard by the housing downturn, according to Chief Executive Officer Frank Blake. His comments helped lift housing stocks, and the Dow Jones Home Construction index rose 0.3 percent.

The Dow Jones industrial average rose 2.71 points, or 0.02 percent, to 13,172.14. The Standard & Poor's 500 Index was down 0.18 point, or 0.01 percent, at 1,403.93. The Nasdaq Composite Index was down 5.54 points, or 0.18 percent, at 3,016.98.

U.S. retail sales rose 0.8 percent in July, the biggest increase since February and better than a forecast 0.3 percent rise. The previous month's numbers were revised to minus 0.7 percent from minus 0.5 percent.

"The next two weeks of data will lead into the annual Fed meeting at Jackson Hole," said Peter Cecchini, managing director at Cantor Fitzgerald & Co in New York, noting that while the headline numbers from the retail sales data exceeded expectations, the previous month's numbers were also revised.

"The revision will likely be ignored, but it produces a print slightly worse than expectations," Cecchini said. He added that the data was important because U.S. Fed Chairman Ben Bernanke has traditionally used the Jackson Hole venue as an opportunity to signal Fed direction.

The better retail sales data dovetailed with strong earnings reports from niche retailers like Estee Lauder .

Reports from smaller retailers also supported the thesis of robust consumer spending that may help lift overall growth.

Michael Kors Holdings Ltd surged 16.5 percent to $49.33 after the apparel company reported higher-than-expected quarterly profit and raised its full-year profit forecast.

Estee Lauder jumped 9.3 percent to $60.13 after the cosmetics and fragrance maker reported a higher-than-expected quarterly profit and forecast more sales growth this year.

But Groupon Inc tumbled 27 percent to $5.51 after the world's largest online provider of daily deals missed quarterly revenue expectations and gave a cautious profit outlook.

The 5.16 billion shares traded on the New York Stock Exchange, the American Stock Exchange and Nasdaq was well below last year's daily average of 7.84 billion.

View the Internet Business Productivity video on YouTube.

Internet business productivity is one of those things that can make or break you online. ?While it is important to use as many "hacks" like this one as you can, it is also important to "just work" -- spending all your time trying out "methods" and "systems" for being productive is NOT the same as actually being productive.

So, watch the above video and get back to work.

Kaspersky Lab urges continuous cyber security education

The architecture of the IT department for most companies has drastically changed over the past few years and with the uptake of mobile devices and virtualisation, security threats have altered. IT security therefore continues to remain a key challenge and priority for IT departments and?this will be the topic of conversation at the IDC IT Security Roadshow to be held?in Johannesburg.

The architecture of the IT department for most companies has drastically changed over the past few years (image: Charlie Fripp)

Worldwide, businesses have had to adapt to the influx of new technological trends such as cloud computing, virtualisation, and Bring Your Own Devices (BYOD).

This has impacted on not only how IT managers approach security but also on how businesses and consumers need to understand the potential security risks associated with the increasing reliance on mobile devices and technological capabilities.

?While security experts tend to focus on future technologies for protecting potential threats, one still needs to be cognisant of incidents that have also happened in the past. By learning from these issues, we are able to build stronger and safer IT security communities for businesses and consumers alike,? says David Jacoby, Senior Security Researcher at Kaspersky Lab and one of the presenters at the IDC IT Security Roadshow.

Although users acknowledge the existence of various online threats, they are generally still reckless when it comes to IT security. In fact, it has been reported that approximately 16% of Microsoft-based laptop users, 50% of Macbook users, and 65% of smartphone users do not use any kind of security software on their devices. This indicates that consumers and businesses still need to be educated on the implications of not taking IT and online security seriously.

?Businesses and consumers are becoming more digitally advanced. As a security company, our responsibility should be to ensure that users are aware of the threats and dangers of the Internet and new technologies, as they continue to emerge. Educating users on such a simple thing as e-mail scams can have a ripple effect on the security landscape of a region,? adds Jacoby.

Knowledge is power and by educating people on the numerous threats and pitfalls the IT landscape has to offer, it is possible to create a society of cyber warriors to fight the fight for safe and secure Internet activity. But it is not just about educating the cyber community ? it is important to continuously update the knowledge available to them to ensure a sustainable approach to IT security.

Risk & Business: Is saying your sorry good risk management for - Yes

Here is the situation, the business insurance broker/agent just realized they messed up. Unfortunately the error caused an uncovered claim against one of their clients. Now what? The first thing an agent is taught to do is call their errors and omissions insurance carrier and say nothing to the client. But is this the right strategy? Many legal scholars and risk managers are starting to suggest different. How about an apology. Yes the simple and effective apology. Now does this dictate liability? Does the apology make the business insurance agent/broker fully responsible? Many courts are now saying not neccessarily. The simple act of saying to someone you have harmed that your sorry does not mean that your at fault. It simply means that you are being human and are genuinely empathetic to the plight of others. Interestingly it is now showing that there are less law suits following an apology. Caution here, always contact your errors and omissions insurance carrier before any communication with your your client after a mess up. They may still require that apology that you want to give to stay in your brain only. ?It is after all still a litigious business insurance environment. ?Until next time be careful out ther and know your risks.

10 Principles to Guide Communicators Today | IABC

Inside organizations today, communication is the greatest challenge for leaders. Employees want to understand the organization?s strategy and how they fit in, but leaders aren?t explaining the reasons behind their decisions. Middle managers especially struggle with how to translate business information and make it relevant to employees and are often seen as the weak link in the organization. What?s more, employees are overwhelmed by information overload. This makes the changing role of communicator more challenging than ever as we help leaders to develop communications that engage employees, according to David Grossman, founder and CEO of the Grossman Group and former director of communications at McDonald?s.

I recently had the opportunity to attend the IABC World Conference in Chicago to hear Grossman and other speakers address the challenges and opportunities of 21st century organizational communications. Grossman shared his top 10 principles for guiding communicators as they work to develop communications to engage the hearts and minds of employees today.

1. Character is the differentiator for leaders and organizations. In today?s rapidly changing business landscape, organizations and leaders must reflect the values principles, beliefs, mission and purpose of the company ? internally and externally. In its report on ?The Authentic Enterprise: Relationships, Values and the Evolution of Corporate Communications,? the Arthur W. Page Society defined four new priorities and skills communications professionals must lead:

? Defining and instilling company values

? Building and managing multi-stakeholder relationships

? Enabling the enterprise with ?new media? skills and tools

? Building and managing trust, in all its dimensions

2. There is no one leadership style. Some leaders are extroverts, others are introverts. Both can be effective communicators, as long as leaders learn to develop ways to bring messages to life in authentic, relatable ways. Communicators can help by developing individual platforms, key messages and elevator speeches for leaders, as well as encouraging them to use storytelling to humanize the company and brand.

3. There is a greater need to imagine and advance a vision. A vision is just a vision until someone acts on it. To make a vision reality, you need the hearts, not just the minds, of employees. Today, the equation is 70% mind, 30% heart. In order to advance a company?s vision, organizations need to change the equation to 60% heart, 40% mind.

4. People really are an organization?s greatest asset. Building brands starts on the inside, and employees play an increasing role in delivering on the brand promise. Employee indifference can negatively impact a brand and sales. Employees are an organization?s greatest ambassadors, so ensure they understand the value proposition.

5. Everyone is a leader. Give employees the opportunity to contribute to the company?s success. Share expectations and hold people accountable. Employees are engaged when they are given responsibility and autonomy and held accountable for results.

6. There is a greater focus on self. To be effective at engaging employees, leaders must be self-aware and understand their level of emotional intelligence ? also referred to as EQ ? and how to relate to others. Help leaders to develop in this area by providing them training and tools.

7. Change is the only constant. Today change happens as a series of changes, not in a linear fashion. Communicating in times of change requires that communicators address proactively what we know, what we don?t know, what we are working on finding out and myths from the grapevine.

8. Employees? fundamental needs won?t change. Employees aren?t likely to understand or buy in to the organization?s vision unless they have answers to the ?me? questions. All employees fundamentally have the same questions: What?s my job? How am I doing? Does anyone care about me? What?s going on? What?s the business strategy? How is the company doing? What are our vision and values? How can I help? Give them the answers and they?ll move from ?me? to ?we.?

9. There is a need for more and better listening individually and systematically. Listening is key to understanding employee perceptions. Communicators need better and more internal data and better listening tools.

10. There is a need for communication systems that are better honed to manage overload and inefficiencies. Communication vehicles like email are effective, but employees are inundated. Some organizations are addressing email overload with email block-outs, mandated email time off and other strategies. Communicators can help by evaluating all communication channels, including email and social media, to ensure they are using them most effectively.

This is the first in a series of reports from the 2012 IABC World Conference by IABC Phoenix Past President Cory Craft.

For another view of David Grossman?s presentation at the IABC World Conference, read communicator Miri Zena McDonald?s post on SmartBlog on Leadership. Miri tweets @mirimcdonald. David Grossman tweets @thoughtpartner.

Plan now to attend the 2013 IABC World Conference in New York City June 23-26.
